This technology is used to measure the effect of a perturbagen on an enzyme activity, which is coupled to a secondary signal. E.g., the activity of a kinase determines the residual pool of ATP, with ATP being its substrate, which in turn determines the rate of a coupled enzyme luciferase, since ATP is one of its substrates too.
