Receptor tyrosine-protein kinase erbB-2 carboxyl terminal fragments (ERBB2 CTF) arise through at least two different mechanisms: proteolytic shedding of the full-length receptor tyrosine-protein kinase erbB-2 extracellular domain and translation of ERBB2 mRNA from internal initiation codons. ERBB2 CTFs resulting from either proteolytic shedding of the extracellular domain or alternative initiation of ERBB2 mRNA from methionine codon 611 contain the transmembrane domain and the intracellular domain (ICD) of the ERBB2 protein and maintain tyrosine kinase activity. Alternative initiation of ERBB2 mRNA from methionine codon 687 results in an ERBB2 CTF that only includes the ICD, which is soluble and localized to the cytoplasm and nucleus; this form does not appear to have tyrosine kinase activity.
