Membrane-Associated Guanylate Kinase Family Gene
Membrane-Associated Guanylate Kinase Family Genes encode MAGUK (Membrane-Associated GUanylate Kinase homolog) proteins containing DHR, SH3, and C-terminal GK-like domains. MAGUKs include Drosophila lethal(1)discs large-1 tumor suppressor protein (dlg1); mammalian Zo-1 tight junction protein; mammalian synaptic proteins that seem to interact with the cytoplasmic tail of NMDA receptor subunits (SAP90/PSD-95, CHAPSYN-110/PSD-93, SAP97/DLG1 and SAP102); 55 kD erythrocyte membrane protein (p55); C. elegans lin-2 protein; rat CASK protein; and human DLG2 and DLG3 proteins. An N-terminal ATP-binding site (P-loop) of Guanylate Kinase is not conserved in the MAGUK GK-like domain; MAGUKs do retain residues known in GK to bind GMP. (NCI) [ ]

Members of membrane-associated guanylate kinase (MAGUK) gene family encode a guanylate kinase (GUK) domain with a catalytically inactive P-loop, which cannot bind ATP. It is thought that MAGUKs use their GUK domain to interact with cytoskeletal proteins and signaling molecules. (HGNC)
